Circular porous cellophane sheets are desinged to fit inside standard petri dishes. The circles have a diameter of 8cm, and they can float on top of media and / or nutritional platforms. The organism of interest can be seeded on top of the cellophane, and can absorb nutrition through the pores. This makes harvesting easier and more efficient.
To sterilize cellophane in an autoclave, you can follow these steps:
- Soak the cellophane in distilled water for five minutes
- Wrap the cellophane in open pore gauze
- Place the cellophane in a pouch for autoclaving
- Set the autoclave profile to 15 minutes at 121°C (250°F)
- Do not activate a dry cycle
- When the cycle is finished, inspect autoclave temperature tape and visually check autoclaved bags
- Bags should be left to cool for several minutes before removing from autoclave
